Research sheds new light on virus associated with developmental delays and deafness

Research sheds new light on virus associated with developmental delays and deafness

Thursday, November 29, 2012

A new study published online in PLOS ONE reveals that primitive human stem cells are resistant to human cytomegalovirus (HCMV), one of the leading prenatal causes of congenital intellectual disability, deafness and deformities worldwide. Researchers from the University of Pittsburgh School of Medicine found that as stem cells and other primitive cells mature into neurons, they become more susceptible to HCMV, which could allow them to find effective treatments for the virus and to prevent its potentially devastating consequences.

"Previous studies have focused on other species and other cell types, but those studies did not evaluate what the cytomegalovirus does to human brain cells," said Vishwajit Nimgaonkar, M.D., Ph.D., professor of psychiatry at the University of Pittsburgh School of Medicine, and senior author of the report. "This study is the first of its kind, and the first to discover that primitive stem cells are actually resistant to HCMV."

Access to cultured human neurons, necessary to understand the pathogenic effects of HCMV, has been limited by difficulties in growing the brain cells in the laboratory. Yet through human-induced pluripotent stem (iPS) cells, researchers were able to overcome this hurdle.

The study authors derived live iPS cells by reprogramming cells called fibroblasts obtained from human skin biopsies. The iPS cells were then induced to mature through several stages into neurons, the primary cells in the brain. The researchers were able to evaluate the patterns of damage caused by HCMV on all these cells.

The research findings suggest:

  • Human iPS cells do not permit a full viral replication cycle, suggesting for the first time that these cells can resist CMV infection
  • CMV infection distorts iPS cell differentiation into neurons, and that may be a mechanism by which infected babies develop impairments of brain maturation and intellectual ability
  • iPS-derived mature neurons are more susceptible to CMV infection and once infected show effects including defective function that have been shown in other animal studies and in other human tissues, and the neurons die a few days after infection lab studies, possibly reflecting the impact of CMV on the human brain

"The findings were quite surprising, but this is only the first in a series of studies on HCMV," added Nimgaonkar. "There is a lot of interest in what we can do to treat the infection, and current work is already underway to screen for new drugs that could be used to fight these viruses."

Between 50 and 80 percent of people in the U.S. have been infected by HCMV by the time they reach 40. Infections are rarely serious, but the virus does not leave the body. CMV is also the most common congenital infection in the U.S., and occurs when a mother contracts CMV during pregnancy and passes the virus to her unborn child. According to the U.S. Centers for Disease Control and Prevention, one of every 150 children is born with CMV infection and one in five of them develops permanent problems, such as intellectual disability, vision and hearing loss, and seizures.

Pitt researchers are collaborating with the Drug Discovery Institute to further understand the cellular system and determine which agents are most effective against HCMV and similar viruses, and which treatments would be safe for human use.

Seven Oscar campaign managers you should know

LOS ANGELES (TheWrap.com) - Notable Oscar campaigners include more mainstream publicity companies like 42West, ID-PR and Ginsberg/Libby, and an array of folks who specialize in foreign films, documentaries and below-the-line talent. Here are some important names to remember:

Tony Angellotti

The Angellotti Company

A 10-year vet of the Miramax wars (though was he never on the Weinstein staff), Angellotti now handles awards for Disney and Pixar's animated films and Universal's live-action ones; this year, that gives him "Les Miserables," "Brave, Frankenweenie" and "This Is 40," among others.

Melody Korenbrot

Block-Korenbrot Public Relations

Korenbrot is a powerhouse in the foreign-language and documentary categories, thanks to BK-PR's longstanding relationship with Sony Pictures Classics ("Amour," "Rust and Bone," "Searching for Sugar Man," "West of Memphis").

Karen Fried

Karen Fried & Associates A former Angellotti Company publicist who went on to work with Lionsgate, Fried serves as the awards publicist for Focus Features, with a 2012 crop that includes "Anna Karenina," "ParaNorman," "Hyde Park on Hudson" and "Promised Land."

Michele Robertson

MRC

With a track record that includes "The Full Monty," "Brokeback Mountain" and "Million Dollar Baby," Robertson handles Oscar campaigns for Warner Bros., which this year means "Argo," "The Hobbit," "The Dark Knight" and "Cloud Atlas," among other high-profile and big-budget contenders.

Jeff Sanderson

Chasen & Company

Taking over from his longtime boss, the late Ronni Chasen, Sanderson has carved out a strong niche representing composers (Mychael Danna), songwriters (Diane Warren) and writers (Ronald Harwood), among others.

Cynthia Swartz

StrategyPR/Consulting

After working countless Oscar campaigns during six years at 42West (and before that 13 years at Miramax), Swartz left that company a year ago to found Strategy. Scott Rudin remains a client, anchoring a roster that this season includes "On the Road," "Zero Dark Thirty" and "Flight."

Murray Weissman

Weissman/Markovitz Communications

A 30-year vet of public relations, Weissman spent 15 Oscar campaigns (and 11 Best Picture nominations) working with Miramax. His company currently works on Paramount's Oscar campaigns.

Longmeadow's Joe Philbin still keeping close New England ties as head coach of Miami Dolphins

FOXBOROUGH ? Joe Philbin will always have deep roots in New England. As a kid growing up in Longmeadow, he spent his Sundays perched in front of the TV watching the AFC East.

Then, when it came time to earn a living, he moved east and started his coaching career at Worcester Polytechnic Institute. A few years later, following various gigs across the country, he ended up at Northeastern as an offensive coordinator, then at Harvard in the same role to ?cut my commute down about 10 minutes,? he said.

After stops with the University of Iowa and the Green Bay Packers, Philbin has made his way back to the AFC East as coach of the Miami Dolphins. It?s a dream job of sorts, but Philbin knows he never would have had the opportunity to host his home-state New England Patriots this weekend if it weren't for the opportunities he found at home with Northeastern and Harvard.

In fact, if the Bay Sate hadn?t taken care of him way back when, Philbin may not be in coaching at all.

?Barry Gallup (the coach at Northeastern at the time), who is the assistant athletic director at Boston College, was nice enough to give me a job,? Philbin said on a conference call from Miami. ?I was unemployed. After going to Ohio University, we went a glamorous 0-11 and didn?t have any job. I had four children, my wife was pregnant and Barry was nice enough to give me a job.?

Philbin made the most of those opportunities, and he still hasn?t forgotten where he came from, even if he?s flown as far south as possible.

Just look at his coaching staff. Fifteen members of it have roots reaching back to New England. Philbin hired nine of them.

Even his top assistants were handpicked from the region. Defensive coordinator Kevin Coyle attended the University of Massachusetts, and offensive coordinator Mike Sherman is a native of Norwood.

Sherman coached with Philbin at Tulane, and later hired him as an assistant offensive line coach with Green Bay in 2003. Their ties go back even further to the 1970s, when Sherman was Philbin?s coach and English teacher at Worcester Academy.

It?s not an accident or a coincidence. Philbin planned it this way.

?When I was fortunate to get this job, I knew it was going to be important to hire, No. 1, excellent coaches, but No. 2, people I could have total trust and faith in. Guys that are loyal,? Philbin said. ?That?s the first and foremost thing.?

So far, the plan has worked well ? somewhat. Philbin isn?t pleased his team is 5-6, and takes particular issue with the fact the Dolphins have committed 21 turnovers despite spending time each day on ball security.

He sees steps in the right direction, but knows things have to improve dramatically over the next few seasons if Miami ever hopes to change the power structure in the division.

?We have some things as a team that we haven?t done yet. This our 11th week together, and we?re playing our 12th game,? Philbin said. ?We have to play better. We?re not necessarily worried about (beating the Patriots in the standings), we?re worried about how well (the Dolphins are) executing in all three phrases. But as far as the standings, that?s not really what we?re worried about.?

But if Philbin?s program grabs hold in Miami and begins harvesting results, perhaps one day the Dolphins will be able to worry about the standings after all.

Healthy Foods
Guilt Free Winter Sips ? Low calorie winter drink ideas

by Leanne Ely, CNC

The holidays are here, and baby, it?s cold outside! Everyone enjoys a cup of something warm but you know the drill?most of the time, these things are calorie bombs in a cup! So how do we navigate this desire for warm beverages vs. the stuff out there that will pack on the pounds and make your thighs expand into New Years?

Winter Sips

Let?s start with some tea, shall we? Is anyone out there a tea drinker? I have English bones, and was raised in a regular tea-sipping home. The only challenge with tea is that it?s hard to break the English tea habit of adding milk or sugar. But I?ve come across a tea that doesn?t need a thing. Good Earth is the brand and the tea that I especially love is the original Sweet and Spicy. It?s a deep red color, with a rich cinnamon taste with sweet tones, leaving the drinker?s tongue infatuated with flavor and subtle sweetness. Give it a try, it?s low in the calorie department and tastes like Christmas in a cup!

For an occasional treat, like after you go cross country skiing to cut down your own Christmas tree (think TREAT!) you can make your own drinks that you would usually buy mixes for: hot chocolate for example.

Hot chocolate is especially easy to make. All you need is unsweetened cocoa baking powder, sugar to taste (be mindful or use a little xylitol insted), a splash of vanilla extract, and a cup of hot milk. In measuring out your own ingredients you don?t have to worry as much about any mysterious additives that tend to increase that calorie count we?re all so cautious of.

Senate Dems rally for Rice against GOP opposition

WASHINGTON (AP) ? Senate Democrats rallied to U.N. Ambassador Susan Rice's defense as Republicans said they were even more troubled by her account of the deadly attack on the U.S. Consulate in Benghazi, Libya, and signaled they would try to scuttle her nomination if President Barack Obama tapped her as the next secretary of state.

"The personal attacks against Ambassador Rice by certain Republican senators have been outrageous and utterly unmoored from facts and reality," said Senate Majority Leader Harry Reid, D-Nev., who called the criticism unfathomable in light of disclosures from the intelligence community.

As congressional Democrats and the Obama administration delivered a full-throated defense of the possible diplomatic nominee, Rice was meeting Wednesday with Republican Sens. Susan Collins of Maine and Bob Corker of Tennessee. Corker is next in line for the top GOP spot on the Senate Foreign Relations Committee.

"We'll see and we're going to sit down and talk to her," Corker told The Associated Press. "She always delivers the party line, the company line, whatever the talking points are. I think most of us hold the secretary of state and secretary of treasury to a whole different level. We understand that they're going to support the administration, but we also want to know that they are independent enough, when administration is off-base, that they are putting pressure. I think that's what worries me most about Rice."

Rice answered questions Tuesday from Sens. John McCain, Lindsey Graham and Kelly Ayotte about her much-maligned explanations about the cause of the September attack in Libya that killed Ambassador Chris Stevens and three other Americans.

At the hour-plus, closed-door session, Rice conceded that her initial account ? that a spontaneous demonstration over an anti-Muslim video triggered the attack ? was wrong, but she insisted she had not been trying to mislead the American people when she made her comments five days later.

"The talking points provided by the intelligence community, and the initial assessment upon which they were based, were incorrect in a key respect: There was no protest or demonstration in Benghazi," Rice said in a statement after the meeting. "While we certainly wish that we had had perfect information just days after the terrorist attack, as is often the case the intelligence assessment has evolved."

She was joined in the meeting by Acting CIA Director Michael Morell.

Rice requested the meeting with the three senators ? her most outspoken critics ? but she failed to mollify them and they indicated they would try to block her nomination.

"We are significantly troubled by many of the answers that we got and some that we didn't get concerning evidence that was leading up to the attack on the consulate," McCain told reporters after a session with Rice that he described as candid.

Said Graham, "Bottom line, I'm more disturbed now than I was before that 16 Sept. explanation." He said in a later interview that Rice went "far beyond the flawed talking points" and should be held accountable.

"I'm more troubled today," said Ayotte, who argued that it was clear in the days after the attack that it was terrorism and not a spontaneous demonstration.

Rice's unusual visit to Capitol Hill ? typically only nominees meet privately with lawmakers ? reflects the Obama administration's campaign for the current front-runner to replace Secretary of State Hillary Rodham Clinton against some strenuous GOP opposition.

The White House remained defiant in its support for Rice, arguing that she was relying on an assessment from the intelligence community and had no responsibility in compiling the information on the cause of the attack. It dismissed what it characterized as a fixation on her national television appearances five days after the raid.

"The focus on, some might say, obsession on comments made on Sunday shows seems to me, and to many, to be misplaced," Obama spokesman Jay Carney told reporters at a White House briefing.

House Democrats, including female members of the Congressional Black Caucus, have suggested that the GOP opposition to Rice is sexist and racist. Senate Democrats, who will increase their advantage to 55-45 in the next Congress, said Rice could win confirmation if Republicans recognize the unfairness of penalizing her for the intelligence community's talking points.

Senate Armed Services Committee Chairman Carl Levin, D-Mich., told reporters "it is so unfair to hold her responsible for something that she didn't produce and which the intelligence community has specifically stood by."

In a statement late Tuesday, McCain, Graham and Ayotte said Morell told them the FBI had removed references to al-Qaida in the talking points to prevent compromising ongoing investigations. Later in the day, the three senators said the CIA contacted them to say Morell misspoke and the CIA had deleted the references.

Microsoft promises to fix random reboots on Windows Phone 8 by next month

Mitt Romney can take some solace in his devastating loss on Nov. 6: at least he won the voters who really count. That's the thesis anyway of top adviser Stuart Stevens, who penned an op-ed in the Washington Post on Wednesday arguing that by winning wealthier and whiter voters, Romney secured the moral victory over Obama. "On Nov. 6, Mitt Romney carried the majority of every economic group except those with less than $50,000 a year in household income," Stevens wrote. "That means he carried the majority of middle-class voters. ...

Stephenie Zamora: Are You Living in Limbo?

Limbo. It's that awkward place between where you are now and where you'll be "soon."

Maybe you're planning to move across the country in the near future. Or your lease is ending in a few months and you're not sure if you'll be able to stay. Perhaps you have to travel for work off and on for the next six months.

So you think to yourself... What's the point in trying to meet someone new and amazing, I'll just be moving soon. Why bother getting that little arts and crafts corner set up in my current place, I may have to box it all up. Why bother taking that class, joining that group or making new friends, I'll just be on the go and away from it all.

The point is that life is precious and way too short to waste in limbo.

Limbo is also the birthplace of unfulfillment, depression, frustration, isolation and general "meh"-ness. When you allow yourself to enter the mental space of limbo, you effectively put living, loving, dreaming, scheming playing, connecting and whatever other wonderful ing-thing you want and desire on hold.

You put life on hold.

And when life is on hold, it slips by fast -- opportunities go unclaimed, amazing love remains undiscovered, friends, laughter, adventures and happiness fade away before you even experience them. All because you're waiting for something that's going to happen "soon."

Your "soon" may be two weeks, two years or two months. Doesn't matter. What matters is that you deserve an amazing, joy-filled life with phenomenal people in it.

2012-11-22-limbo.png

So how do you stop living in limbo?

Start living in the moment.

One of the most transformational lessons I've learned in my life thus far is how to live in this exact moment, rather than getting lost in the mess of thoughts, negative chatter, worry, fear or drama in my head.

Think about it for a moment. (Yes, I'm giving you temporary permission to go in your head.) Instead of putting yourself out there, falling in love, making amazing friends, having fun and exciting life experiences, you're probably sitting at home or work thinking about how you'd love to be doing those things, but can't, since you have this change happening "soon."

Here's the truth, you can have those things. So what if you're only here for two more months? Get yourself out there and do all the things you want to do! Date (maybe even fall in love), meet new people (find the bestie you've always dreamed of), take that art class (so you'll miss the final show... at least you'll have made some art)!

Start showing up to every moment of every single day fully present and engaged in what you're doing or who you're with. Stop thinking about how things "have to be" because of something happening later. Be here now for this moment and have the most amazing experience you can.

Stop being afraid of what you can't control or foresee.

Unless you're some kind of amazing psychic, you don't know what's going to happen with anything, so stop assuming that you do.

"They won't want to hang out with someone who's moving soon."

"No one would ever want to date me seriously the way I have to travel."

"I'm not going to be able to stay here, anyway."

You don't know that. It's not possible for you to know that, especially when other people are involved. So stop assuming! Your "soon" is in the future, not right now. For all you know, you could meet a kindred spirit that you connect so deeply with you remain best friends via text, email and Skype for the rest of your lives. You could meet the man of your dreams who, by the time "soon" arrives, is willing to make things work. Or, your landlord will let you stay in your place and you just wasted a couple months not doing crafts because you assumed you'd have to move.

Stop being afraid or "realistic" about things... just live your life.

Remember that you can't control everything.

I strongly believe that if something's meant to happen, it will happen, whether you're ready for it or not. So if you're supposed to connect with someone specific, come across a certain opportunity or be guided in a certain direction, it's going to happen. Trust that everything happens for a reason and start moving with the flow of life.

Don't try so hard to maintain "control" of your life and your plan. When you do, you miss out on amazing things. Unexpected job offers, the person you're supposed to spend your life with, that best friend you've always wanted,

Just because you planned to move, change jobs, get married or whatever else, doesn't mean that's what's supposed to happen for you. If life presents you with an opportunity and your intuition says "hell yes!" you'd better listen up and act accordingly. Trying to control every little thing just causes unnecessary stress, frustration and anxiety. Loosen up your grip, begin listening to and trusting your intuition, and start to enjoy the amazing journey that is life.

Most importantly, stop wasting time in limbo!

Take action now!

If you feel like you're living in limbo, grab a notebook or journal and answer the following questions:

  • Why am I so afraid/hesitant to do the things I want to do?
  • How would it feel to do them anyway and really start living and enjoying my life in the present moment?
  • Where can I relax my need for control and start moving with the flow of life?

Then... close your eyes, take a deep breath and re-engage in your life, in this moment (and every single moment after that) fully and completely.

Greek bankruptcy averted ? for now | The Salt Lake Tribune

Managing Director of the International Monetary Fund Christine Lagarde, second right, speaks with the media as she arrives for a meeting of eurogroup finance ministers at the EU Council building in Brussels on Monday, Nov. 26, 2012. Eurozone finance ministers are set to meet in Brussels on Monday to discuss the next installment of bailout money for debt-laden Greece. (AP Photo/Virginia Mayo)

Athens, Greece ? European and global financial leaders have agreed to release $57 billion in critical loans to Greece and provide billions in additional debt relief in order to help the country stabilize its ailing economy.

After three weeks of negotiations, Greece?s euro partners and the International Monetary Fund agreed early Tuesday morning to release the loans in four installments beginning next month. The leaders also settled on a raft of measures ? including a debt buyback program and an interest rate cut on loans ? that will reduce the country?s debts by about $52 billion.

Greek Prime Minister Antonis Samaras hailed the agreement in Brussels as a victory. "Yesterday, a very grey, a very dark time for Greece ended definitively," he said in a televised address to the nation, adding that the agreement "managed to ensure us remaining in the euro."

But the country will still face years of economic pain as austerity measures agreed to as part of the bailout package are implemented.

Most stock markets in Europe were modestly higher on the news out of Brussels with the Stoxx 50 index of leading European shares closing up 0.2 percent. Meanwhile the euro gave up earlier gains to trade 0.4 percent lower at $1.2941. The interest rate charged on Greece?s benchmark 10-year bonds, an indicator of investor confidence in a country?s finances, fell 0.2 percentage points to 14.47 percent on the news of the debt deal.

"There remains the potential for this deal to fall apart in the medium term as there are a lot of moving parts and it is a long way away from the permanent fix that the IMF had been insisting upon," said Gary Jenkins, managing director of Swordfish Research.

"It is just one more big kick of the can down the road."

For three years, Greece has been struggling to convince markets as well as its creditors that it can get a grip on its public finances, which had spiraled out of control. The country is predicted to enter its sixth year of recession and is weighed down by an unemployment rate of 25 percent.

The so-called troika of the European Central Bank, IMF and the European Commission has twice agreed to bail out Greece, pledging a total of $310 billion in rescue loans ? of which the country has received about $194 billion so far. In return for its bailout loans, Greece has had to impose several rounds of austerity measures and submit its economy to scrutiny.

Without the bailout money, the country would be facing bankruptcy and a possible forced exit from the 17-country eurozone. This would have potentially chaotic repercussions for the world economy.

story continues below

Nonetheless, the spending cuts and reforms insisted on by the troika have been painful. Ordinary Greeks are struggling to make ends meet as wages have been cut and taxes increased. The country is routinely shut down as strike after strike is called in protest of yet more austerity. Meanwhile, extreme political views on both the left and right are enjoying increased popularity.

The eurogroup and IMF agreed to release in December $43 billion in loans originally scheduled for June. The remainder will be issued in three installments in the first quarter of 2013. The money will be used to help recapitalize Greece?s struggling banking industry and pay back suppliers, including its pharmacists which have gone for months without any payment from the Greek state welfare system.

Greek Finance Minister Yannis Stournaras said the deal was "very important for it keeps Greece in the euro, offers it a significant opportunity to exit the vicious cycle of recession and over-indebtedness, and contributes to its debt reduction."

But Germany?s finance minister, Wolfgang Schaeuble, warned that Greece still has to stick to its side of the bargain for the bailout loans and debt relief to work.

"We can do what we want, from the IMF to the eurogroup, if Greece itself doesn?t implement the necessary, difficult reforms and adjustment measures step by step, then it?s a mission impossible," he told reporters in Berlin.

The meeting in Brussels was the third time in the three weeks that eurozone finance ministers had tried to hammer out a deal on the next installment of bailout money for Greece.

The main aim of the bailout program is to right Greece?s economy and get it to a point where it can independently raise money on the debt markets once the bailout loans start to run out at the end of 2014. It has been clear for months that the country is far from achieving that goal. Greece?s debt levels are expected to hit 190 percent of its annual economic output next year? some $470 billion. The talks have centered on trying to get Greece back on the path to sustainability by reducing the country?s debt load.

Afghans protest against death sentence for soldier who killed French troops

[unable to retrieve full-text content]

Source: www.hizb.org.uk --- Tuesday, November 27, 2012
Students protest at death sentence for Abdul Saboor when western Troops are not executed for civilian deaths Hundreds of Afghan students have demanded the release of an Afghan soldier sentenced to death for killing five French soldiers, in a rowdy protest that was a reminder of hostility to western forces and the central government in many parts of Afghanistan. Abdul Saboor was given the death penalty by a military court for opening fire on French forces during a joint operation in eastern Kapisa province in January. The deaths contributed to France?s decision to withdraw its Troops early, and highlighted the growing problem of insider attacks by Afghan police and soldiers against the foreign soldiers training them. The shootings have become one of the biggest challenges for Nato commanders in Afghanistan. This year so far more than 60 foreigners have been killed in 45 such attacks, around 15% of military deaths in the country. The rising trend has pushed the United States to restrict contacts with Afghan Troops and raised questions about the long-term future of the training mission. Afghan forces have suffered similar or higher losses to attackers from inside their own ranks, and have stepped up screening procedures to try to limit what they see as treacherous attacks. Facebook Gifts Now Lets You Buy iTunes Credit For Friends And Recommend What They Should Purchase

Facebook iTunes GiftsFacebook and Apple deepened their alliance today, as Facebook Gifts is starting to sell iTunes digital gift certificates. Upon purchase, Facebook users can suggest what music, video, or apps their friends should spend their $10, $15, $25, or $50 credit on. By adding one of the world's most popular presents to its Gifts store, Facebook Gifts could boost sales and split the 30% margins with Apple. What the actual revenue split is remains a secret. We do know there's no shipping costs for either partner to pay for, and Apple typically taxes content and app creators a 30% to be sold in iTunes.

Turn Everyday Objects Into Fitness Equipment - Health X Pert Articles

Exercise equipment is great at the gym, but if you don?t want to shell out the money for at-home workouts, you can just make your own. There are a bunch of products already in your home that can double as exercise equipment. See if you have these three everyday items for getting your sweat on in your home gym. Instead of: handheld weights Use: canned tomatoes. Soup or other dense canned foods may not be skinny in the middle like dumbbells, but the ridges on the side will help keep your grip. Choose two of the same weight to work your arms. How: use canned food in place of any handheld weight exercise you would do for your shoulders, biceps, or triceps. If you have options in your pantry, use the lightest cans for your shoulders and triceps while using the heavy set for biceps. Instead of: resistance band. Use: the tie from your bathrobe. Many exercise classes are swapping out resistance bands for cloth karate ropes. A bathrobe tie works just as well as its black-belt counterpart. How: this waist tie is great for stretching your legs. Use it while lying on your back to stretch your hamstrings, IT band, or calves by simply extending one leg into the air and hooking the strap around the sole of your foot. This is also a unique weapon for ab work. Try ab exercises where your legs are suspended out in front of you and your back is off the ground. You can loop the belt around one or both of your legs for more support and to take the strain out of your neck. Keep reading for our third idea.

Built to Last: Climate Data Ensure Oil Supply Route

David Miller, Louisiana Department of Transportation and Development

I?m David Miller and I was the project manager for the original LA-1 project that you see behind you here. When I was in high school, I could walk out there several hundred yards. Now you have to have a boat to get out there. And the sea level rise that?s occurring, that is something we had to take into account when we built this structure.

The data we received and used from NOAA to make this thing last 75 years was critical. Through a cooperative effort with NOAA early on in the project, we were able to gather data and information through the GPS technology that told us what was happening. And we could then go back and verify what we were seeing. Frankly, what we found out was a little scary. We?re losing almost a half an inch a year because of sea level rise*.

So we had to take that into account looking forward. I?m very confident the information that we gathered and that we used in the design and planning of this structure is going to give us a 75 year design life. That?s important because just down the road there is Port Fourchon that supplies a tremendous amount of the offshore oil industry, specifically the deep water. It was very important to have that information. It was very important to know that we would have confidence that it would work.

*Editor?s note: This rate is for relative sea level rise, which includes both rising global ocean levels and the impact of subsidence, which is the settling and sinking of the ground over time.

'Two and a Half Men' star calls show 'filth'

By Jordan Zakarin and Lesley Goldberg, The Hollywood Reporter

At this point, it might be best to turn "Two and a Half Men"?into a reality show.?Less than two years removed from?Charlie Sheen's infamous meltdown that saw him removed from the CBS?Chuck Lorre?comedy, the show's young star,?Angus T. Jones?-- the half-man in the show's title -- has blasted the series as "filth" and suggested that people should stop watching.

More from THR: Miley Cyrus to Romance Jake on 'Two and a Half Men'

"Jake from 'Two and a Half Men'?means nothing. He is a non-existent character," Jones says in a video for the?Forerunner Christian Church, which is based in Fremont, Calif. "If you watch 'Two and a Half Men,' please stop watching 'Two and a Half Men.' I'm on 'Two and a Half Men'?and I don't want to be on it. Please stop watching it and filling your head with filth. People say it?s just entertainment. Do some research on the effects of television and your brain, and I promise you you?ll have a decision to make when it comes to television, especially with what you watch."

Jones continues with the testimonial, intimating that the show is part of the plan of "the enemy" -- presumably Satan. He adds, "If I am doing any harm, I don't want to be here. I don't want to be contributing to the enemy's plan ... You cannot be a true God-fearing person and be on a television show like that. I know I can't. I'm not OK with what I'm learning, what the bible says and being on that television show."

Photos from THR: Broadcast TV's Returning Shows for 2012-13

Jones, 19,?gave a testimony?in October at the Voice of Prophecy in Los Angeles, where he attends services.

Jones earns about $350,000 per episode; in May he received a salary bump -- along with co-stars?Jon Cryer?and?Ashton Kutcher?-- when?Men?was renewed for a 10th season.

Jones' comments mark the latest brouhaha for "Men," which in addition to the Sheen debacle also saw former showrunner?Lee Aronsohn?step down after he came under fire for comments he made to?THR?about female-driven shows at a Toronto screenwriting conference.

"Two and a Half Men"?producer Warner Bros. Television declined comment.

What do you think of Jones' statements? Tell us on Facebook.

Ruth Whippman: How Corporate America Is Turning Into a Cult and Why It's Harming the American Employee

I used to work for a large organization. One afternoon, all the staff in my department were herded into a giant conference room, and informed that due to budget cuts within the year over half of us present would lose our jobs. Fresh from a leadership course at the University of Corporate Doublethink, our bushy-tailed manager "reframed" the concept for us. "Don't see this as a negative" he advised. "We like to see this as us empowering you to take on new opportunities."

Over the last decade or so, a growing disconnect has developed between the bizarre and almost cult-like rhetoric and practices that companies use with their staff, and the increasingly grim reality of being an employee in modern day Corporate America.

For example, anyone stumbling upon the Walmart careers website might be forgiven for thinking that Google had malfunctioned and directed them instead to the Scientologists, whose recruitment site features a collection of testimonials virtually indistinguishable in their tone of robotic devotion.

Over at "Walmart People," Lois Givens, Personnel Manager at store number 992 assures us: "If you live your whole entire life according to the Walmart culture and three basic beliefs, life becomes a lot easier." Shana Bailey, Director of Store Operations emotes: "To this day, I continue to grow and learn, and the Walmart family is always there for me every step of the way," while Patricia Graham of the Distribution Centers adds: " Walmart is my Life (capitalization her own). When I think about it, it's amazing how many aspects of my life are touched and made better by Walmart."

For those of us whose lives have yet to be touched by the transformative miracle of the out of town superstore, these testimonials may seem a little excessive. But these are not rogue sycophants in the company ranks. According to Michael Bergdahl, former "Director of People" for Walmart's Headquarters, and now an international speaker and consultant on the company's practices, the creation of "cult-like commitment" and devotion to the so called "Walmart Way" among its employees is the explicit aim of the company's intensive staff training program for new recruits. And lest employees forget what they have learned on the course, loyalty is reinforced daily in stores with the "Walmart cheer," a compulsory devotional chant.

It's not just Walmart. Corporate songs are common, "reframing" disappointment for those whose lives have taken the road marked "middle manager" rather than the one marked "rockstar." "We built this Starbucks on Heart and Soul," "Blue Box Values Are the Best, I Work For American Express" and perhaps most uplifting, Big Four accountancy firm Ernst and Young's reworking of gospel anthem "Oh Happy Day" which replaced the lyrics: "When Jesus washed my sins away" with the competing message "When Ernst and Young showed me a better way."

Once they have whipped their staff into a frenzy of corporate sycophancy, employers are increasingly expecting them to turn their whole selves over to the company's message and aims, and are employing a range of sophisticated psychological and practical techniques to achieve this.

Silicon Valley firms encourage their employees to work ever longer hours by turning offices into playgrounds, complete with slides, graffiti walls and foosball tables. Free dinner and on site dry cleaners, dentists and hairdressers are standard ways of ensuring that staff never leave the building. Facebook made the perfect metaphor for the overworked American employee into a reality with their flagship 'Insanity Wolf' treadmill desk, which allows employees to keep running to a never materialising destination while they continue to work. I met (a rather tired looking) Facebook employee recently who was thrilled to be provided with pizza and "allowed" to work overnight on a company project of his choice at no extra pay.

The boundaries between self and company are further blurred as employers increasingly encroach onto personal territory in order to keep their staff firmly in the workplace and not outside it. Biotech firm Genetech will organize babysitters for its staff rather than have them take time off to stay at home with a sick child. Consulting firm Deloitte offers backup care for an elderly parent or grandparent. Human Resources philosophers have ditched the mantra "work life balance" for the more sinister (and employer friendly) "work life integration."

Corporations now aim to identify and satisfy employees' emotional needs as well as their financial ones. Major white collar employers such as UBS, American Express, KPMG and the law firm Goodwin Proctor have instituted "happiness training" for staff, drawing on a combination of psychological research and ancient religious traditions. Other employers sponsor their staff to go on courses such as the hugely popular Landmark Forum series, several full days in a group setting in which bathroom breaks are discouraged and participants are invited to share personally disturbing life events such as relationship problems and family breakdowns with the group and then go through a signature process to "heal" these issues and find lasting peace.

But while loyalty to an employer and pride in one's work can be important planks of self esteem, the increasing merge of self and company is a cultural shift that poisonously favors the employer over the employee. The more the relationship is characterized by unswerving devotion rather than genuine bargaining power, the more employers are able to encourage staff to work harder and longer, and the more they are able to suppress any meaningful dissent.

It's a strategy that's working. For all this cultish rhetoric, the power balance between employer and worker has not been loaded further in the employer's favor since the second world war. Executive pay and profits have skyrocketed while wages have stagnated and vast numbers of blue-collar workers are living below the poverty line, dependent on food stamps and government assistance just to get by. These two facts are not unrelated -- a JPMorgan economist calculated that the majority of increased corporate profits between 2000 and 2007 were the result of "reductions in wages and benefits."

Despite all this talk of "empowerment" American workers have now lost any real bargaining power they once had with their employers to change these conditions. Union representation, with its promise of better wages and benefits for workers, has reached a record low. The Walmart workers' strike comes after decades of methodical discouraging of union membership by their employers. They are not the only ones. Among others, Starbucks ("we're called partners because it's not just a job, it's our passion") lost in Federal Court after being accused of illegal union busting efforts and firing baristas for union activities.

White collar workers are affected by this trend too, with punishingly long hours at no extra pay now the norm. The average American employee spends a staggering month longer on the job each year than their 1969 counterpart and vast swathes of companies including Bank of America, Starbucks and IBM have been implicated in lawsuits for failing to pay their office staff appropriate overtime. Easy for bosses to dismiss as a necessary pulling together in tough economic times, this is a trend that long predates the recession and one from which those at the very top have only benefited.

It would seem that the more Corporate America demands cult-like devotion from its employees the worse it is able to treat them, and the less likely they are to complain. Yet given that the vast majority of Americans stand to lose rather than gain from this type of relationship, it is worth injecting a bit of healthy scepticism back into our assessment of our employers' motives. Now that really would be empowering.

No winner of huge Powerball prize ? it hits $425 million

People lined up for Powerball tickets Saturday, the fourth largest Powerball in history. NBC's Thanh Troung reports.

By NBC News staff and wire services

DES MOINES, Iowa -- No one won a huge Powerball prize on Saturday night, and lottery officials said the jackpot would jump to a record $425 million for the next drawing -- and possibly higher.

The winning numbers were?22-32-37-44-50 with Powerball 34, according to the Powerball lottery's website.?According the Powerball website, there were 10 winners of $1 million and one winner of $2 million.

Iowa Lottery spokeswoman Mary Neubauer said?the jackpot for Wednesday's drawing could go even higher than the estimated $425 million because sales pick up in the days before record drawings.?

Saturday's jackpot was the fourth-largest in the game's history.?The previous top Powerball prize was $365 million, won in 2006 by eight ConAgra Foods workers on a single ticket in Lincoln, Neb.

The prospect of a big payoff had pulled in Black Friday shoppers in many cities.

Chicago resident Clyde Gadlin, 65, emerged from the bustle of holiday shoppers on Chicago's Michigan Avenue, to stop in at a 7-Eleven to buy his daily batch of Lottery tickets, including Powerball.

For him, the game is a chance to dream ? a single winner's cash payout would be nearly $213 million before taxes ? and he tries not to let the long odds burst his bubble.

Lottery officials say they're unsure what effect Thanksgiving and beginning of Christmas shopping season will have on sales, which normally pick up in the days before high-dollar drawings.

Gadlin said that if he won, he would return to his grandfather's farm in Heidelberg, Miss., where he spent part of his childhood.

"I would go down there again and probably do a little bit of farming," he said, recalling the roaming deer and 380 acres of potatoes, corn, watermelons and sugar cane. Gadlin hasn't been there for more than 20 years.

Since Powerball tickets doubled in price to $2 in January, the number of tickets sold has decreased, but the sales revenue has made up for it, increasing by about 35 percent, said Norm Lingle, chairman of the Powerball board of directors.

And as the price went up, so did the jackpots, enticing thousands across the country to play.

"Christmas is coming and $325 million would come in handy," said Tim Abel, 63, who was buying a Powerball ticket at New York's Port Authority Bus Terminal. The Broadway stagehand said he usually plays whenever the jackpot goes over $100 million.

Recent Powerball winners include an Iowa couple that won a whopping $202 million on Sept. 26. A week later, a Delaware resident picked all six numbers for a $50 million payday.

Cashiers at a DeliMart convenience store in Iowa City said they expect a last-minute rush Saturday. The store sells $200 to $300 worth of Powerball tickets on an average day, but assistant manager Scott Falkenhan said he anticipates that will more than double.

Falkenhan, 32, was planning to buy his own ticket after his shift ended.

"I'd buy a new truck and a new boat," he said. "Then I'd go on my own fishing tour."

The current jackpot is the fourth largest in the game's history, with chances of winning at about one in 175 million. Many dream of winning it big and hope they will be that lucky someone with the golden ticket. NBC's Mark Potter reports.

Lingle, who is also the executive director of the South Dakota Lottery, says this weekend will be "telling."

"To my knowledge we've never had a large jackpot run like this fall over a major holiday," he said.

Frank Scorvino of Toms River, N.J., is in charge of buying lottery tickets ? including Powerball ? for a group of about a dozen fellow construction workers. If they win, he said, most of them would purchase new houses and cars, and some might go on a cruise.

South Bend, Ind., resident Dameon Smith says he'd retire from his job at the Hummer plant in Mishawaka, Ind. He said he'd share with family, friends and people he doesn't know.

Chad Robinson, 41, a chef at a Cleveland restaurant, had an option during his break Friday: Put down $2 on Powerball game or go next door to Ohio's first casino. He bet on the lottery, saying that much cash would change his life "drastically."

"I figure I'll make a lot of people happy with it, not just myself, spread the love and live my life out ? parents, loved ones, kids, co-workers, charities," he said.

Terry Fowler, 50, of Conneaut, Ohio, was visiting family in Tennessee for the holiday and stopped in a gas station in Brentwood on Friday morning to buy Powerball tickets. He's a regular player.

"I want to see more than one person hit it so they can share the wealth," said Fowler, a sales representative with a food service company. "I don't think any one person needs $325 million. If 7-10 people hit that, they will live like kings."

Bolt, Felix named IAAF World Athletes of the Year

BARCELONA, Spain (AP) ? Usain Bolt won the IAAF World Athlete of the Year award for the fourth time Saturday after defending his 100 and 200 meter gold medals at last summer's Olympic Games in London.

The Jamaican sprinter, who beat out American hurdler Aries Merritt and Kenyan runner David Rudisha, had previously won the award in 2008, 2009, and 2011.

"For me this is a great honor to win a fourth time. I really worked hard and I was really focused this year. This season was one of my toughest. I had my ups and downs, even though we don't like to talk about them," said Bolt, who thanked his coach and the fans at the Olympics.

"The London games were magnificent. Fans were there in the stadium from 10 in the morning. I had never seen anything like that."

Sprinter Allyson Felix of the United States took home the women's award.

In 2003, Bolt and Felix won the Rising Star awards for the most promising young athletes of the year.

"Nine years ago I was here with Allyson, now I am back, that means I am getting old, which is slightly worrying," said the 26-year-old Bolt. "But I will continue to work hard."

Felix won the 200-meter gold medal in London after settling for silver four years earlier in Beijing.

"My biggest defeats have come at the Olympic Games. After Beijing I was devastated, I had worked as hard as possible but I came up short," she said. "But I decided to rededicate myself even harder and it was a blessing in disguise. I was able to work harder than ever before and finally it all came together."

The other finalists for the women's award were New Zealand shot putter Valerie Adams and British heptathlete Jessica Ennis ? both gold medalists in London.

Merritt won the Inspiration Award for his season, which included a gold medal and a world record in the 110-meter hurdles.

Rudisha, who was named Athlete of the Year in 2010, won the Performance of the Year award for his record-setting gold medal run in the 800 meters.

Javelin thrower Keshorn Walcott of Trinidad and Bahamas sprinter Anthonique Strachan won the Rising Star awards.

The awards were announced on the final day of the IAAF's 100th anniversary celebration.

The current jackpot is the fourth largest in the game's history, with chances of winning at about one in 175 million. Many dream of winning it big and hope they will be that lucky someone with the golden ticket. NBC's Mark Potter reports.

DES MOINES, Iowa --?Black Friday shoppers in many cities briefly detoured into lottery retailers, drawn off task by the prospects of winning a $325 million Powerball jackpot on Saturday night ? the fourth-largest in the game's history.

If Gadlin wins, he said he'd return to his grandfather's farm in Heidelberg, Miss., where he spent part of his childhood.

"I would go down there again and probably do a little bit of farming," he said, recalling the roaming deer and 380 acres of potatoes, corn, watermelons and sugar cane. Gadlin hasn't been there for more than 20 years.

And if he isn't successful this time, it's likely he'll have another shot at a record-breaking pot of cash.

Roxie Breece, an assistant manager at a Cenex convenience store in Ogallala, Neb., said clerks have sold far more Powerball tickets than usual over the past week.

"Tomorrow's going to be a nightmare for us," she said. "With everybody out shopping and the drawing on Saturday, we'll be really busy."
